* All injections included Freund’s adjuvant, other adjuvants are available at an additional cost; four injections are normally recommended and at least 2 hens are recommended.
** One pre-immune egg and about 20 immune eggs are normally collected from each hen. The immune eggs will be collected 1 weeks after the second immunization.
***A total of about 60 days is normally recommended. Extended period can be requested with additional cost of $1 per day. Daily maintenance includes care, feeding and veterinary care if necessary.
NOTE: We will not accept radioactive, toxic, or infectious antigens.

ANTIBODY PURIFICATION
1. Antibody Purification (AP-1)- Total IgY (about 50mg/egg): IgY will be purified to about 90% purity and protein concentration will be determined. $20/egg for individual egg, $10/egg for pooled eggs. Total cost is about $360 (12 x $20 = $240 for individual eggs, 12 x $10 = $120 for pooled eggs) for 500-1000mg IgY.
2. Antibody Purification (AP-2): This service includes antibody purification using Protein-A affinity chromatography. Price: $ 65 for 5-10 mg IgG (human IgG, mouse IgG, rabbit IgG and bovine IgG), $85 for 10-30 mg, $105 for 30-50 mg, $125 for 50-100 mg, $650 per g and $500 per g for > 2 g, For goat IgG, add 15% more.
3. Antibody Purification (AP-3): This service includes antibody purification using Protein-G affinity chromatography. Price: $ 85 for 10-20 mg IgG, $105 for 20-40 mg, $135 for 40-60 mg, $175 for 60-100 mg, $925 per g, $750 per g for >2 g.
4. Antibody Purification (AP-4): This service includes antibody purification using Protein-L affinity chromatography. Price: $ 90 for 10-20 mg IgG, $105 for 20-40 mg, $135 for 40-60 mg, $165 for 60-100 mg, and $750 per g.
5. Antibody Purification (AP-5): This service includes antibody purification using Protein-LA affinity chromatography. Price: $ 95 for 10-15 mg IgG, $115 for 20-40 mg, $135 for 40-60 mg, and $180 for 60-100 mg, and $950 per g.
6. Affinity Purification (AP-6): This service includes antibody purification using protein, peptide or other molecules made affinity chromatography. Price: $600 for yield from 2-10mg. We need about 5-10mg free peptide, ideally peptide with a cystein at C or N terminal, or 5-10mg protein (or fusion).
SDS-PAGE of Antigens
Antigen Purification - about 3 mg antigen needed for four injections of 2-3 hens. Customer provides information on protein concentration and buffer conditions. The purity of target protein must be >30%. Cost: $160 per antigen.
Conjugation of up to 10 mg of antigen to two conjugates (KLH or BSA) - Cost: $150
Conjugation of same antigen to an additional carrier – Cost: $100
ANTIBODY TITRATION BY ELISA
TITRATION ASSAY IN 96 WELL MICROPLATE 50 - 100ug antigen needed per plate for coating and protein concentration must be provided. Need to know buffer conditions
$200 for first microplate, $150 for each additional microplate
This is a four-lane-assays. Need 100ug antigen, approximate molecular weight of protein of interest, protein concentration and buffer conditions. Cost: $200 for first four lanes, $100 for each additional four lanes.
We provide low background secondary Abs such as bovine anti-IgY biotin and anti-IgY HRP (see Conjugates Section). These Abs are suitable for ELISA, Western Blot and IHC/ICC. However, we also provide biotinylation service. Cost: $150/5mg.
We help customers to find and design the best antigenic peptides as immunogens. In general, 10-20 aa peptides are selected as antigens. Cost: $30 per aa for 10-20mg MAP (multiple antigenic peptide with lysine branches) peptides.
WESTERN BLOT ANALYSIS PROTOCOL
Western Blot Analysis Protocol
1. After transferring proteins from SDS-PAGE to nitrocellulose membrane, block the membrane with blocking buffer (5% nonfat milk in TBS) for 1 hour at RT, or overnight in 4oC.
2. Dilute Abs accordingly in Abs dilution buffer (3% nonfat milk in TBST (TBS plus 0.1% Tween 20)) and add the Abs in each blot.
3. Shake at RT for at least 2 hour or keep in 4oC overnight.
4. Empty the solution and wash at least 3X10 minutes with TBST with shaking.
5. Dilute conjugate in 3% nonfat milk/TBST (Genetel Anti-IgY AP 1:3000) and incubate blots with the conjugate for 1 hour, shaking.
6. Empty the solution and wash at least 4X10 minutes each with TBST.
7. Wash 1X 10 minutes with TBS.
8. Lay the membrane in a flat container and do color development using AP substrate.
Buffer
TBS:
NaCl 8g KCl 0.2g
Trizma Base 1.21g D.I. Water 1L
Adjust pH to 8.6 with 1M HCl
Blocking Buffer: TBS + 5% Nonfat Dry Milk.
TBST:
1L TBS
1 ml Tween-20 (Polyoxyethylene-Sorbitan Monolaurate)
Abs Dilution Buffer: TBST + 3% Nonfat Dry Milk
Note: Due to the nature of IgY sticky property, you may need to increase Tween-20 in your TBST.
